Enzyme cold adaptation

Proteins are affected by temperature. Low temperatures affect protein function and protein folding. We are examining sequence changes in enzymes from cold and temperate environments to find out if there is some change in structure that might cause an increase in enzyme activity in the cold or affect the efficiency of protein folding at low temperatures.

Current work focuses on a model enzyme, lactate dehydrogenase, from a group of related fish. Experimental work includes:


  • Protein isolation and purification
  • Kinetic characterization
  • Protein stability measurement
  • Protein expression in E. coli and the production of specific mutants
  • Crystallization and structure determination
  • Protein folding assessment
